A group of experienced genomic sequencing researchers coordinated by Professor Davide Cacchiarelli at the Telethon Institute of Genetics and Medicine in Pozzuoli, Tigem, determined the complete sequence of the viral genome in 115 positive swabs for SARS-CoV-2 performed in the last months in the region.

Among these samples it was also possible to analyze 6 swabs recently obtained at the Capodichino airport in people from London: among these the presence of the so-called "English variant" of SARS-Cov2 was found, on which many scientific aspects still remain to be clarify to assess their actual impact on public health. In the remaining samples analyzed, a total of 8 different variants were identified, all belonging to the "type B", widespread in Europe.

The study, the results of which have been deposited in Gisaid (the world reference database for sequences of viral genomes), therefore provides a first map of the Sars-CoV2 variants present in Campania and underlines the importance of viral genome analysis for the characterization of the evolution of the pandemic. "At TIGEM we have developed a technology that now allows us to analyze, with high precision and reliability, the viral genome in a high number of swabs per day. 26/12/2020 - Six passengers from the United Kingdom tested positive for the so-called English variant of Covid-19 because they have the S gene mutations typical of this variant. The news emerged following the work carried out by the researchers of the Experimental Zooprophylactic Institute of the South, in synergy with Tigem and Ceinge. Further studies are underway in close collaboration with researchers from the Spallanzani Institute.

The study is part of the Gencovid project, which started from the Campania Region in the first months of the pandemic with the aim of studying the virus in depth through the timeliness of sampling on the territory, the identification of outbreaks and the phylogenetic reconstruction of the virus and its spread. . A scientific boarding of the best Campania research institutions which, thanks to the synergy of the researchers, has allowed the Campania Region to identify and sequence about 2000 samples in the various phases of the pandemic, data that will be published in a few days on the international Gisaid platform and made available of the international scientific community. Gencovid was born from the Izsm which, with 400,000 swabs analyzed in the laboratories of Portici and with specialized operating units, allowed targeted and immediate interventions in the management of emergencies. Once the RNA was extracted and prepared, the Italian excellence Tigem for molecular biology took care of the sequencing, a further aliquot of sample, in collaboration with the CEINGE researchers, was also used to isolate the viral strains and experimentally test in vitro, on cellular models, the efficacy of different molecules with potential inhibitory activity of the replicative capacity of the virus.

The English variant

The "English variant" of Sars-CoV-2, the Covid-19 virus, was found in Veneto (3 cases), Campania (6 cases) and Abruzzo (one case). Two cases had been ascertained yesterday in Puglia . The variant, as is known, is more contagious, not more serious, and is called "English" because it was found for the first time in Great Britain. The variant is characterized by a combination of deletions (absence of small pieces of the viral genome) and mutations in the S protein (spikes) that make it unique within the panorama of the many variants circulating in the world.
